Native DNA.

A partnership programme for modified ssDNA. Synthesis services for linear and circular ssDNA. Existing constructs for research.

Modified ssDNA partnership programme.

Internal chemistry at kilobase length

Partner with Moligo to develop long ssDNA with modified nucleotides distributed throughout the strand. Our modification technology is available to selected partners through a co-development programme with an agreed development scope, not available as fee-for-service.

Together, we assess chemistry feasibility, select the base channel and substitution fraction, and agree characterisation and application testing for the construct. Substitution is global, with dose control per base channel.

Linear ssDNA

Custom linear ssDNA
up to 10 kb.

Custom HDR donor templates

Encode your sequence in long ssDNA molecules up to 10 kb. Every construct is scar-free and sequence-verified, released at 100% Sanger alignment to your target sequence.

We synthesise both donor templates for non-viral gene editing and DNA vectors with ITRs for episomal expression. Catalogue constructs target the TRAC and AAVS1 loci for T cell and CAR-T work. Enzymatic synthesis handles sequence features that constrain other routes, including repeats, high GC content and strong secondary structures. We review each construct before quoting and tell you what we can make.

Circular ssDNA

Custom circular ssDNA
up to 10 kb.

Circular donors for non-viral gene insertion and episomal expression

Combine a custom donor sequence with a closed circular format. Explore topology as a design choice for your gene-insertion or episomal expression workflow.

Moligo’s collaboration with Cellectis evaluated circular versus linear donors in HSPCs. The study supports the circular format under its tested conditions.

Read each result in context.

Sanger alignment is a sequence-identity check, not a per-molecule error-rate measurement. A gel band does not by itself establish a numerical purity percentage or a residual double-stranded DNA detection limit.

Modified DNA needs additional characterisation.

Modification technology is available to selected partners. Within each co-development programme, we agree how to measure incorporated chemistry and composition, and which format and purity assays are appropriate.
